With the Green Deal the EU aims to achieve a circular economy, restore biodiversity and reduce environmental pollution. As a part of the Green Deal a 'one-substance one-assessment' (OS-OA) approach for chemicals has been proposed. The registration and risk assessment of chemicals on the European market is currently fragmented across different legal frameworks, dependent on the chemical's use. In this review, we analysed the five main European chemical registration frameworks and their risk assessment procedures for the freshwater environment, covering 1) medicines for human use, 2) veterinary medicines, 3) pesticides, 4) biocides and 5) industrial chemicals. Overall, the function of the current frameworks is similar, but important differences exist between the frameworks' environmental protection goals and risk assessment strategies. These differences result in inconsistent assessment outcomes for similar chemicals. Chemicals are also registered under multiple frameworks due to their multiple uses, and chemicals which are not approved under one framework are in some instances allowed on the market under other frameworks. In contrast, an OS-OA will require a uniform hazard assessment between all different frameworks. In addition, we show that across frameworks the industrial chemicals are the least hazardous for the freshwater environment (median PNEC of 2.60E-2 mg/L), whilst biocides are the most toxic following current regulatory assessment schemes (median PNEC of 1.82E-4 mg/L). Finally, in order to facilitate a successful move towards a OS-OA approach we recommend a) harmonisation of environmental protection goals and risk assessment strategies, b) that emission, use and production data should be made publicly available and that data sharing becomes a priority, and c) an alignment of the criteria used to classify problematic substances.

The potential water demand for fracturing fluids along with the possible flowback and produced water production is assessed for the Dutch Posidonia shale. Total water demand estimated for 25 years of the field development using historic data from the U.S. plays varies between 12.2 and 36.9 Mm3. The maximal annual water consumption of 0.95–2.88 Mm3 is expected in the peak years of shale gas production. These figures are much lower than the availability of any potential water sources, which include drinking water, fresh and brackish groundwater, river water, effluents of wastewater treatment plants (WWTP) and sea water. River water is considered the most promising water source for fracturing fluids in the Dutch Posidonia shale based on its availability (>6·104 Mm3/year) and quality (only bacterial composition needs to be controlled). Total wastewater production for the whole period of the field development is estimated between 6.6 and 48.0 Mm3. Wastewater recycling can cover significant part of the source water demand for fracturing fluid. However, high mineral content of the wastewater as well as temporal and spatial discrepancies between wastewater production and water demand will form obstacles for wastewater recycling. The assessment framework developed in this study may be applied for other shale gas fields with high uncertainties regarding subsurface properties, connate formation water characteristics and future legislative framework.

A plethora of in vitro bioassays is developed in the context of chemical risk assessment and clinical diagnostics to test effects on different biological processes. Such assays can also be implemented in effect-based monitoring (EBM) of (drinking) water quality in parallel to chemical analyses. EBM can provide insight in the risks for environmental and human health associated with exposure to (unknown) complex low-level mixtures of micropollutants, which fits in the risk-based approach that was recently introduced in the European Drinking Water Directive. Some challenges remain, in particular related to selection and interpretation of bioassays. For water quality assessment, carcinogenesis, adverse effects on reproduction and development, effects on xenobiotic metabolism, modulation of hormone systems, DNA reactivity and adaptive stress responses are considered as the most relevant toxicological endpoints. An evaluation procedure of the applicability and performance of in vitro bioassays for water quality monitoring based on existing information has been developed, which can be expanded with guidelines for experimental evaluations. Besides, a methodology for the interpretation of in vitro monitoring data is required, since the sensitivity of specific in vitro bioassays in combination with sample concentration may lead to responses of chemicals (far) below exposure concentrations that are relevant for human health effects. Different approaches are proposed to derive effect-based trigger values (EBTs), including EBTs based on 1) relative ecotoxicity potency, 2) health-based threshold values for chronic exposure in humans and kinetics of reference chemicals, and 3) read-across from (drinking) water guideline values. EBTs need to be chosen carefully in order to be sufficiently but not over-conservative to indicate potential health effects. An increase in the volume and diversity of chemicals is one of the main drivers of this deterioration, of which biodiversity loss is a telling indicator. In response to this situation, in October 2020, a chemicals strategy for sustainability (CSS) was published in the EU. The CSS is the first regional framework aiming to address chemical pollution in a holistic manner. The CSS covers the complete lifecycle of a chemical, including the design of better substances and remediation options, to remove chemicals from the environment. The strategy contains terms, such as a "toxic-free environment," for which no clear definition exists, potentially hampering the implementation of the CSS. In this paper, a definition for a "toxic-free environment" is proposed on the basis of a survey and a discussion held at the 2020 SETAC Europe Annual Meeting. In addition, key issues that are absent from the CSS but are considered to be key for the realization of a toxic-free environment are identified. To achieve the policy goals, it is recommended to align the definition of risk across the different chemical legislations, to establish a platform for open data and data sharing, and to increase the utility and use of novel scientific findings in policymaking, through the development of a strong science to regulation feedback mechanism and vice versa. The paper concludes that environmental scientists have the tools to address the key challenges presented in the CSS. However, an extra step is needed by both policymakers and scientists to develop methods, processes and tools, to increase the robustness and transparency of deliberation processes, and the utility of science. We identify uncertainties and knowledge gaps of chemical risk assessment related to unconventional drillings and propose adaptations. We discuss how chemical risk assessment in the context of unconventional oil and gas (UO&G) activities differs from conventional chemical risk assessment and the implications for existing legislation. A UO&G suspect list of 1,386 chemicals that might be expected in the UO&G water samples was prepared which can be used for LC-HRMS suspect screening. We actualize information on reported concentrations in UO&G-related water. Most information relates to shale gas operations, followed by coal-bed methane, while only little is available for tight gas and conventional gas. The limited research on conventional oil and gas recovery hampers comparison whether risks related to unconventional activities are in fact higher than those related to conventional activities. No study analyzed the whole cycle from fracturing fluid, flowback and produced water, and surface water and groundwater. Generally target screening has been used, probably missing contaminants of concern. Almost half of the organic compounds analyzed in surface water and groundwater exceed TTC values, so further risk assessment is needed, and risks cannot be waived. No specific exposure scenarios toward groundwater aquifers exist for UO&G-related activities. Human errors in various stages of the life cycle of UO&G production play an important role in the exposure. Neither at the international level nor at the US federal and the EU levels, specific regulations for UO&G-related activities are in place to protect environmental and human health. UO&G activities are mostly regulated through general environmental, spatial planning, and mining legislation.

Water authorities and drinking water companies are challenged with the question if, where and how to abate contaminants of emerging concern in the urban water cycle. The most effective strategy under given conditions is often unclear to these stakeholders as it requires insight into several aspects of the contaminants such as sources, properties, and mitigation options. Furthermore the various parties in the urban water cycle are not always aware of each other's requirements and priorities. Processes to set priorities and come to agreements are lacking, hampering the articulation and implementation of possible solutions. To support decision makers with this task, a decision support system was developed to serve as a point of departure for getting the relevant stakeholders together and finding common ground. The decision support system was iteratively developed in stages. Stakeholders were interviewed and a decision support system prototype developed. Subsequently, this prototype was evaluated by the stakeholders and adjusted accordingly. The iterative process lead to a final system focused on the management of contaminants of emerging concern within the urban water cycle, from wastewater, surface water and groundwater to drinking water, that suggests mitigation methods beyond technical solutions. Possible wastewater and drinking water treatment techniques in combination with decentralised and non-technical methods were taken into account in an integrated way. The system contains background information on contaminants of emerging concern such as physical/chemical characteristics, toxicity and legislative frameworks, water cycle entrance pathways and a database with associated possible mitigation methods. Monitoring data can be uploaded to assess environmental and human health risks in a specific water system. The developed system was received with great interest by potential users, and implemented in an international water cycle network.

At the same time, governmental agencies demand risk reduction to prohibit any often unknown adverse effects, and industrial parties demand smart approaches to reduce uncertainties. Responsible research and innovation (RRI) is therefore a central theme in policy making. Risk analysis and technology assessment, together referred to as "RATA," can provide a basis to assess human, environmental, and societal risks of new technological developments during the various stages of technological development. This assessment can help both governmental authorities and innovative industry to move forward in a sustainable manner. Here we describe the developed procedures and products and our experiences to bring RATA in practice within a large Dutch nanotechnology consortium. This is an example of how to put responsible innovation in practice as an integrated part of a research program, how to increase awareness of RATA, and how to help technology developers perform and use RATA. Abstract
Present evaluations of chemical pollution in European surface and groundwater bodies focus on problem description and chemical classification of water quality. Surprisingly, relatively low attention has been paid to solutions of chemical pollution problems when those are encountered. Based on evaluations of current practices and available approaches, we suggest that water quality protection, monitoring, assessment and management of chemical pollution can be improved by implementing an early-stage exploration of the 'solution space'. This follows from the innovative paradigm of solution-focused risk assessment, which was developed to improve the utility of risk assessments. The 'solution space' is defined as the set of potential activities that can be considered to protect or restore the water quality against hazards posed by chemical pollution. When using the paradigm, upfront exploration of solution options and selecting options that would be feasible given the local pollution context would result in comparative risk assessment outcomes. The comparative outcomes are useful for selecting optimal measures against chemical pollution for management prioritization and planning. It is recommended to apply the solution-focused risk assessment paradigm to improve the chemical pollution information for river basin management planning. To operationalize this, the present paper describes a still-growing database and strategy to find and select technical abatement and/or non-technical solution options for chemical pollution of surface waters. The solutions database and strategy can be applied to help prevent and reduce water quality problems. Various case studies show that implementing these can be effective, and how solution scenarios can be evaluated for their efficacy by comparative exposure and effect assessment.

Abstract
Background
The number of chemical parameters included in monitoring programs of water utilities increased in the last decade. In accordance with the European Drinking Water Directive, utilities aim at a tailored risk-based monitoring (RBM) program. Here, such a RBM program was developed for the largest Dutch water utility, mostly using groundwater as a source. Data from target analyses and high-resolution mass spectrometry-based suspect screening was used to cluster the different source waters. Targets were prioritized based on (preliminary) drinking water guideline values or the threshold of toxicological concern. Suspects were prioritized for further identity confirmation based on semi-quantitative occurrence concentrations combined with in vitro toxicity information. Finally, a RBM program was suggested for each cluster of source waters.

Results
Out of 731 target chemicals, 153 were detected at least once over a 5-year period. Roughly 10% of the detected non-target screening features matched to suspects. 108 source waters were clustered into 7 clusters. Source waters with low numbers and concentrations of organic chemicals were located in areas with all land-use types, while clusters of source waters with higher numbers of chemicals were related to infiltrated surface water. For perfluorinated chemicals, 25 suspects matched features detected in source waters and 7 features detected in drinking water. For the target chemicals, simple treatment showed the lowest and sorption-based techniques relatively high removal efficiencies. The chemical composition of all drinking waters related to non-contaminated source waters. (Preliminary) guideline values were available for 45 of the retrieved target chemicals, and used for prioritization for monitoring frequencies. These chemicals individually posed no appreciable concern to human health. Suspects were prioritized for further identity confirmation based on semi-quantitative occurrence in produced water, detection frequencies and information on toxic potency. Once confirmed and assessed as relevant, the suspects could be added to target monitoring.

Conclusion
This approach provided a feasible workflow for RBM of target chemicals for clusters of groundwater sources, connected to a feed of new relevant chemicals based on suspect screening.
