Aflatoxins (AFS) are toxic and carcinogenic fungal metabolites. Aflatoxin B1 is the most toxic and has been classified as a Group I carcinogen by the International Agency for Research on Cancer (IARC). Samples of imported rice were analyzed for their AFS content. Finley ground rice subsamples were extracted with water/methanol (100:150 v/v) followed by purification with Immunoaffinity columns (IAC). AFS purified from extracts were determined with RP-HPLC-FLD using post column electrochemical derivatization with a Kobra Cell. Concentrations of aflatoxin B1 and total AFS in test rice samples were ≤0.123 and ≤2.58 µg/kg, respectively. Tween 80 improved recoveries (86 and 106%) of aflatoxin B1 and aflatoxin G1 from brown rice. Recoveries of Aflatoxin B2 and aflatoxin G2 were substantially reduced (non-detected to 27%) by Tween 80 used in IAC cleanup of brown rice extracts. Visible dense growth of Aspergillus parasiticus (food isolate) occurred at 25 °C but higher aflatoxin B1amounts (23.9–39.3 µg/kg) accumulated when the mold grew at 37 °C in rice seeds stored for three weeks. It could be concluded that levels of aflatoxin B1 and total AFS in rice samples were within the permissible amounts of the EU and other international legislations.

Aflatoxin contamination is a serious concern given their hepatotoxic properties and their widespread occurrence during cultivation, harvest, drying, storage, transit and distribution. Preharvest infection by A. flavus is the major cause of aflatoxin contamination in peanut. Its prevention is a complicated task that requires a series of intervention strategies to be merged with traditional farming practices. The International Crops Research Institute for the Semi-Arid Tropics (ICRISAT) and its partners have developed an integrated approach to mitigate A. flavus infestation and aflatoxin contamination by combining: (i) host plant resistance, (ii) soil amendments with lime and organic supplements to enhance water holding capacity, plant vigor and seed health, (iii) timely harvesting and postharvest drying methods, (iv) the use of antagonistic biocontrol agents, such as Trichoderma and Pseudomonads, and (v) awareness campaigns and training courses to disseminate technology to the end-users. This approach can successfully reduce aflatoxin contamination in peanuts in West and Central Africa. This approach is simple, economical and suitable for subsistence farming conditions, but also can be scaled up for use on commercial farms in developing countries in Africa and Asia. ; European Union ; Peer Review

Aflatoxins (AF) are a group of fungal toxins that contaminate foods and feeds with adverse health impact on human beings and animals. A total of sixty three (63) samples of Groundnut kernels from three (3) Agricultural zones of Sokoto State, were analysed for the presence of Aflatoxins B1 (AFB1), B2 (AFB2), G1 (AFG1) and G2 (AFG2) using High Performance Liquid Chromatography (HPLC). The Aflatoxin was detected in 82.5%, the total Aflatoxin concentration ranged between 0.9-646.0 μg/kg. All four types of AF were detected from all samples, out of the total samples analysed 38.10% contained AF levels above the Nigerian (20 μg/kg) limits for AF, while 66.7% samples had AF concentrations above the European Union maximum tolerance level of 2μg/kg for AFB1 and Total Aflatoxin(Tot AF) of 4μg/kg. The result shows that, there were significant (P > 0.05) differences between the samples analysed in the state, as regards to AFB1. The results also revealed that Groundnut kernel in the study area have high frequency of contamination (82.5%). However, awareness on danger of Aflatoxin contamination by all stakeholders is the key in effective management of the problem.Relevant quality control units must be reactivated to assess the quality of the Groundnut kernels from which other products are made.

Improving both quality and quantity of food available is a pressing need especially when one eighth of the world's population consumes less energy than is required for maintenance and is exposed to contaminated food, both of which lead to greater susceptibility to diseases. The Pakistani population depends heavily on milk for nutritional needs and 10% of household income is spent on milk. This commodity requires continuous monitoring and care from its site of production by smallholder dairy producers through to urban consumers along tradition milk marketing chains. Feed ingredients used as concentrate feed to enhance milk production are often contaminated with mycotoxins, which, after ingestion, are transferred into milk. Aflatoxins can contribute to the causation of liver cancers, immune system disorders, and growth-related issues in children. Moreover, deaths in both humans and animals have also been reported after ingestion of aflatoxin-contaminated food. Studies have shown contamination of food and feed ingredients with mycotoxins, especially aflatoxins. This review places the dairy industry into context, summarizes how milk and milk products are contaminated with aflatoxins, and discusses the present legislative regulation of milk quality implemented in Pakistan. There is a need to eliminate fungus-susceptible animal feed ingredients, which are the source of mycotoxins so prevalent in the milk marketed to the consumer in Pakistan.

Kenya has had several outbreaks of aflatoxicosis affecting humans and pets. The aim of this study was to compare aflatoxin levels in cereals, the staple diet of Kenyans, in Nandi where aflatoxicosis has not been reported despite being a maize growing area. Maize, sorghum and millet were sampled from households and also from markets serving various villages in the selected sub-locations (408 samples). The samples were tested for total aflatoxin contamination using cELISA. Households in the study sub-locations depended on homegrown grains than market sourced. Sixty seven point nine percent (72/106), 73.3% (44/60) and 65.7% (67/102) of maize samples collected from Laboret, Kilibwoni and Chepkongony were contaminated with aflatoxins ranging between 0.17-5.3 parts per billion (ppb). Ninety two point nine percent (13/14), 100% (9/9) and 87.5% (14/16) of millet samples from Laboret, Kilibwoni and Chepkongony were positive for aflatoxin at a range of 0.14-6.4 ppb. Fifty percent (9/18), 36.4% (8/22) and 27.3% (6/22) of sorghum samples from Laboret, Kilibwoni and Chepkongony, respectively were contaminated with aflatoxins beyond Kenya Bureau of Standards (KEBS) maximum tolerable limits of 10 ppb. To manage aflatoxin contamination of the cereals in Nandi, the county government needs to step up awareness creation of the dangers posed by chronic aflatoxin exposure to households through cereals and promote good Agricultural practices.

Processed infant food samples (ready to eat) were analyzed for aflatoxins contamination using Reverse Phase High Performance Liquid Chromatography (RP-HPLC) with fluorescent detection. A solvent mixture of acetonitrile-water was used for the extraction followed by immunoaffinity clean-up to enhance sensitivity of the method. The limit of detection (LOD) (0.01–0.02 ng/g) and limit of quantification (LOQ) (0.02 ng/g) was established for aflatoxins based on signal to noise ratio of 3:1 and 10:1, respectively. The processed food samples tested, 38% were contaminated with four types of aflatoxins, i.e., AFB1 (0.02–1.24 μg/kg), AFB2 (0.02–0.37 μg/kg), AFG1 (0.25–2.7 μg/kg) and AFG2 (0.21–1.3 μg/kg). In addition, the results showed that 21% of the processed foods intended for infants contained AFB1 levels higher than the European Union permissible limits (0.1 μg/kg), while all of those intended for adult consumption had aflatoxin contamination levels within the permitted limits.

The aflatoxin contamination of food products is a prime parameter to analyze the quality of food. The present work specifically involves the determination of aflatoxin content of food through chromatographic technique. Pakistan Food Authority and European Union Regulations were followed for quantitative analysis of AFTs in pulses and spices. The samples were collected from different regions of Punjab including Sialkot, Narowal, Gujranwala and Gujrat. The AFTs detected in 120 samples, as per detection limits defined by regulatory authorities i.e. 50 ppb for spices and 4 ppb for pulses through Thin Layer Chromatography (TLC). Maximum AFTs was observed in (spices) red chili samples from Gujranwala city (55.5 ppb), black pepper samples from Narowal city (65.9 ppb), coriander samples from Shakar Garh (67.9 ppb), cumin samples from Daska (63.9) and aniseed samples from Sambrial (52.5 ppb). On the other hand, maximum AFTs for pulses was observed in split chick pea sample from Lahore (11.2 ppb), lentils samples from Kingra (8.6 ppb) and black gram beans from Gujrat Fuwara Chowk (15.4 ppb). This study provides awareness to the public for AFTs contamination in different food commodities.

Exposure to unsafe level of aflatoxin in early life may lead to growth faltering. However, the extent of contamination of breast milk and complementary foods is poorly examined. We determined aflatoxin M(1) (AFM(1)) and B(1) (AFB(1)) contamination of human breast milk and cereal‐based cooked complementary foods, respectively, among households having children 6–23 months of age in Sidama zone, southern Ethiopia. Data were collected through two cross‐sectional surveys implemented in the wet (n = 180) and dry (n = 180) seasons. Eligible households (n = 360) were recruited from three agroecological zones (lowland, midland and highland, each with sample size of 120) using a multistage sampling technique. AFB(1) and AFM(1) levels were determined using enzyme‐linked immunosorbent assay. Mann–Whitney U and Kruskal–Wallis tests were performed to compare aflatoxin levels between seasons and across the agroecological zones. Among 360 breast milk samples tested, 64.4% had detectable AFM(1) and 5.3% exceeded the 0.025 parts per billion (ppb) limit set by the European Union for infant milk. The median AFM(1) in the lowlands was significantly higher than in the other agroecological settings (P < 0.001). By season, AFM(1) was higher in breast milk samples collected in the dry season (P = 0.041). AFB(1) was detected in 96.4% of the food samples tested, and 95.0% had concentration exceeding the permissible European Union limit of 0.1 ppb. The median AFB(1) was significantly higher in the lowland (P = 0.002), but there was no difference between the seasons (P = 0.386). The study indicated that, in southern Ethiopia, foods intended for infants are heavily contaminated with AFB(1). Contamination of breast milk is also a significant health concern.

Aflatoxin contamination of maize is a serious food safety problem worldwide. Despite the widespread consumption of maize in Nigeria, there is limited data on aflatoxin contents of maize vended in open markets in Ondo state, Nigeria. A total of 140 maize samples randomly purchased from major markets in four locations in Ondo state, were screened for total aflatoxins using an ELISA method. Exposure and health risk assessments were performed for the maize consumers by the deterministic and Margin of exposure (MOE) approaches, respectively. About 99% of the maize were contaminated with total aflatoxins (range: 0.65–265 µg/kg; mean: 125.9 µg/kg). Aflatoxin levels exceeding the 4 µg/kg set by the European Union for total aflatoxins were found in 88% of the maize whilst more than one half contained at least 100 µg/kg aflatoxins. The average probable daily intake values were 830, 332 and 138 ng/kg bw/day for the average children, adolescent and adult populations, respectively. Consequently, MOEs for the respective populations were 0.20, 0.51 and 1.23, suggesting a high level of health risk for consumers of maize vended in open markets in Ondo state due to high aflatoxin levels. Maize farmers and households in Ondo state need urgent aflatoxin mitigation interventions.

Background: Aflatoxins are a group of mycotoxins that have been associated with hepatic damage and cancer. Aflatoxins B1 and B2 are secondary metabolites produced by fungi Aspergillus. These toxins can be found in a variety of commodities, especially in maize, and have been studied around the world due to their effects in human health. The Latin American population is especially exposed to aflatoxins given that maize products can be found in traditional diets all over the continent. Interestingly, in Mexico, chronic hepatic diseases and cirrhosis are leading causes of death in adult population.Methods: In order to observe the effect of physical variables like temperature and humidity, this study was conducted collecting samples in four different seasons, in two communities in the State of San Luis Potosi, in Mexico. The content of aflatoxins in tortillas was measured using immunoaffinity columns prior to HPLC-FLD analysis.Findings: Results showed that 18% of samples exceeded the Mexican limits for AFB1; whereas, 26% of the samples exceeded the limits of the European Union for AFB1. The AFB1 was detected in 80% of samples in one site and higher concentrations were found in samples collected during fall and winter seasons.Conclusions: Lack of control in storing practices is the principal cause for the contamination of maize. Considering that maize products are part of the staple diet of Mexican population, our results show that AFB1 detection has to be declared a public health priority. Detection and prevention of aflatoxins through a surveillance program, may avoid chronic health effects.

In addition, the farmers' level of knowledge of the causes and consequences of contamination and the measures for prevention were also examined by administering questionnaires to a cross section of 150 farmers. The results showed the presence of aflatoxins in all samples, with levels ranging from 1.6 to 2,270 μg/kg. In 68% of all samples, total aflatoxin contamination was above 4 μg/kg, the maximum tolerable level set by the European Union. Farmers ranked high humidity, improper storage practices, and poor soils as potential causes of aflatoxin contamination and changes in color, smell, and taste, and difficulty in selling crops as consequences. They identified crop management practices as the most effective way to control contamination. The results also revealed that most farmers apply preharvest crop management practices as a means of controlling contamination. More educated households were more knowledgeable about aflatoxins. Female‐headed and married households were less likely to be willing to pay for aflatoxin control. About 28% of farmers claimed to be willing to allocate resources to seed intervention while a smaller proportion agreed to pay for training and information services. The result further suggests that an adoption of pre‐ and postharvest technologies together with awareness creation is still required to reduce aflatoxin contamination in the country.
