Europa - und in geringem Maße auch Nordamerika - steht, wie allgemein bekannt, einer "asiatischen Herausforderung" gegenüber. Dieser Umstand lädt zu Vergleichen ein: Was ist es, das Asien bzw. große Teile davon , so schnell vorankommen läßt, während Europa zurückfällt? Auch besteht, mit Blick auf die Tatsache, daß die Erfolge in Asien mit anderen Mitteln erreicht worden sind, weiterhin der Eindruck, daß das Beispiel, das die (west-) europäische Integration nach dem zweiten Weltkrieg gegeben hat (also das "Modell Europa"), in Asien gescheitert ist. This study analyses how and why developing countries internalise and enforce (or not) policies promoted and prescribed through the EU's governance approach in Zambia. The motivation of this research is the arguments made in the scant literature on policy transfer, which predominantly privileges the policy entrepreneur such as the EU and lack of structured analysis on the EU's governance approach. First, the limited documentation on developing countries as policy recipients assumes that they accept and implement policies due to fear of consequences or in return for financial incentives. Second, the literature supposes that a policy transfer process is smooth, automatic and uncontested. Third, the EU introduced a 'new' governance approach in its development cooperation, which was accompanied by an incentive tranche. The approach assumed that recipients would commit and reform governance areas in return for financial incentives. This tranche faced resistance and partners such as Zambia never implemented most commitments. Despite this, there are limited studies on this approach. This qualitative study goes beyond explaining the spread of the EU's governance reforms and explores the adoption and implementation process of the changes promoted and prescribed in Zambia. A case study method is employed, and governance-related programmes are examined. In particular, the study analyses the Support to the Electoral Cycle Management project and Access to Justice programme, through which electoral processes and judicial and law reforms, amongst a few, were adopted. In addition to analysing the two cases, this study relied on interviews with Zambian academics, bureaucrats, civil servants, consultants and practitioners as additional first evidence on why Zambian actors would adopt and enforce EU governance reforms domestically or not. These semi-structured interviews also allowed the study to obtain descriptions of the world of the interviewee concerning interpreting the meaning of the described phenomena. While following the core tenets of social constructivism, the study makes use of the policy transfer framework to conceptualise the interactions, implementation and opposition of EU reforms in Zambia from the policy recipient's perspective. An analysis of these case studies and interviews showed that the policy transfer process is not self-perpetuating as recipients of policy are not passive actors. While the EU transfers reforms of policies, the Zambian Government and national institutions can decide whether to internalise or oppose the reforms. Therefore, the fear of consequence or increased development aid does not necessarily ensure enforcement of policies by the recipients. Instead there are factors within the adoption mechanisms (conditionalities, incentives, policy transfer entrepreneurs, multileveling and lesson drawing) that determine whether the policy recipient will copy and harmonise the transferred policies at the domestic level or not. These factors include inclusion and ownership; social appropriateness; social, financial and trade benefits; political will of domestic actors to reform and the nature of the partnership between the recipient and the entrepreneur. The study makes some empirical and theoretical contributions. The study highlights what the EU's development cooperation could be doing to ensure efficiency and effectiveness from both partners, amongst other contributions. This study also illustrates that material factors are not the driving force for adopting and implementing policies at the domestic level. The Zambian Government and national institutions are active in deciding the direction and tenor of governance. The adoption and implementation of transferred policies are dependent on the presence of social, economic, political, institutional and policy factors in the adoption pathways. Furthermore, the study finds that policy transfer and a social constructivist approach as conceptual and theoretical worldview offer a plausible explanation for the internalisation process beyond what the conventional International Relations theories on the EU can do. ; Diese Studie erläutert wie und warum die Entwicklungsländer, die im Rahmen des Europäische Union (EU) 'Governance Approach' in Sambia geförderte und vorgeschriebene Verfahrensweisen verinnerlichen und umsetzen (oder nicht). Die Motivation dieser Forschung sind die Fehlen einer strukturierten Analyse des 'Governance Approach' der EU und die Lücken und Grenzen in der existierenden Literatur zum Thema Politik-Transfer die überwiegend den Fokus auf Politikunternehmer wie die EU legen. Erstens geht die begrenzte Literatur über Entwicklungsländer als Empfänger der Politik davon aus, dass diese aus Angst vor Konsequenzen oder als Gegenleistung für finanzielle Anreize Richtlinien akzeptieren und umsetzen. Zweitens geht die Literatur davon aus, dass ein Politikübertragungsprozess reibungslos, automatisch und unangefochten stattfindet. Schließlich führte die EU einen neuen 'Governance Approach' in ihrer Entwicklungszusammenarbeit ein, begleitet von einem finanziellen Anreizsystem, 'governance incentive tranche'. Dieses Anreizsystem rief Widerstand hervor und Partner wie bspw. Sambia haben die meisten Verpflichtungen nicht übernommen. Abgesehen davon gibt es wenige Studien über diesen Ansatz. Diese qualitative Studie geht über die Erklärung der Verbreitung der 'Governance' Reformen der EU hinaus und untersucht den Prozess der Übernahme und Umsetzung der in Sambia angekündigten und vorgeschriebenen Änderungen. Es wird eine Fallstudienmethode angewendet und es werden 'Governance' bezogene Programme untersucht. In der Studie werden insbesondere das Projekt 'Support to the Electoral Cycle Management' und das Programm 'Access to Justice' analysiert, durch die unter anderem Wahlprozesse und Justiz- und Gesetzesreformen verabschiedet wurden. Die Studie folgt den Kernprinzipien des sozialen Konstruktivismus und nutzt die Rahmenbedingungen für den Politiktransfer um die Umsetzung und die Ablehnung von EU-Reformen in Sambia aus der Perspektive des politischen Empfängers zu konzeptualisieren. Neben der Analyse der beiden Fallstudien stützte sich diese Studie auf Interviews mit sambischen Akademikern, Bürokraten, Beamten, Beratern und Praktikern. Dies ist ein zusätzlicher erster Hinweis darauf, warum sambische Akteure die Reformen der EU-Governance im Inland annehmen oder durchsetzen. Diese semi-strukturierten Interviews ermöglichten es der Studie auch, Beschreibungen der Welt des Befragten bezüglich der Interpretation der Bedeutung der beschriebenen Phänomene zu erhalten. Eine Analyse dieser Fallstudien und Interviews hat gezeigt, dass der Prozess des Politik- Transfers nicht selbstbeständig ist, da die Empfänger von Governance-Politik keine passiven Akteure sind. Während die EU Politik Reformen transferiert, können die sambische Regierung und die nationalen Institutionen entscheiden, ob sie diese Reformen verinnerlichen oder ablehnen werden. Daher gewährleistet die Furcht vor Konsequenzen oder vermehrte Entwicklungshilfe nicht unbedingt die Durchsetzung von Richtlinien. Stattdessen Faktoren wie Inklusion und 'ownership'; soziale Angemessenheit; soziale, finanzielle und handelspolitische Vorteile; eine Initiative der nationalen Akteure zur Reform und zur Art der Partnerschaft zwischen Politikempfänger und Politikunternehmer, die im Rahmen der Adoptionsmechanismen vorhanden ist (Konditionalitäten, Anreize, 'Policy Transfer Entrepreneurs', 'Multileveling' und 'Lesson Drawing'), gewährleistet das Kopieren, Nachahmen und Harmonisieren der übertragenen Politiken auf nationaler Ebene. Das Fehlen dieser Faktoren in den Mechanismen führt zur Nichtübertragung. Die Studie leistet einige empirische und theoretische Beiträge. Die Studie hebt hervor, was die Entwicklungszusammenarbeit der EU tun könnte, um neben anderen Beiträgen Effizienz und Wirksamkeit beider Partner zu gewährleisten. Diese Studie zeigt auch, dass materielle Faktoren nicht die treibende Kraft für die Annahme und Umsetzung von Maßnahmen auf nationaler Ebene sind. Die sambische Regierung und die nationalen Institutionen bestimmen aktiv die Richtung und die Dauer der Regierungsführung. Die Annahme und Umsetzung der übertragenen Maßnahmen hängt von der Präsenz sozialer, wirtschaftlicher, politischer, institutioneller und politischer Faktoren ab, unter anderem in dem Prozess der Annahme. Darüber hinaus stellt die Studie fest, dass der 'Politik-Transfer' und ein sozial konstruktivistischer Ansatz als konzeptionelles und theoretisches Weltbild eine plausible Erklärung für den Internalisierungsprozess darstellen, die über das hinausgeht, was herkömmliche Internationale Beziehung-Theorien über die EU leisten können.

Das wissenschaftlich anspruchsvolle und praktisch gerade vor dem Hintergrund aktueller Entwicklungen wirtschaftlich bedeutsame Thema befasst sich mit der Frage, welche Schutzrechte für dreidimensionale Produktgestaltungen in Betracht kommen und in welchem Verhältnis sie zueinander stehen. Eine Produktgestaltung kann z.B. zunächst Schutz nach dem Designrecht genießen und nach dessen Ablauf wettbewerblichen Nachahmungsschutz oder Markenschutz anstreben. Die Frage der sukzessiven Gewährung verschiedener Schutzrechte für ein und dieselbe Produktgestaltung und damit deren "Überlappung" und Kumulation ist gerade mit Blick auf den Schutz dreidimensionaler Marken durch das auf Dauer verlängerbare Markenrecht von großer Brisanz, weil er zu einer Perpetuierung und Monopolisierung des Schutzes führen kann. Die Arbeit gliedert sich grob in fünf Teile. Im 1. Teil werden im Rahmen der Einleitung die Problemstellung, die Zielsetzung und der Ablauf der Arbeit skizziert. Im 2. Inhaltsangabe:Einleitung: Der enorme Anstieg von Auslandsdirektinvestitionen (ADI) durch multinationale Unternehmen (MNU) in den letzten zwanzig Jahren ist eines der deutlichsten Zeichen der Globalisierung der Weltwirtschaft. Auslandsdirektinvestitionen sind zum wichtigsten Bestandteil von privaten Kapitalflüssen zwischen Ländern geworden. Durch die immense, wirtschaftliche Integration weltweit, die viele Handelsbarrieren und Hindernisse für internationale Investitionen beseitigt hat, entstanden immer mehr und größere multinationale Unternehmen. Laut Weltbank wuchs das Volumen von Auslandsdirektinvestitionen von ca. US$ 55 Mrd. im Jahre 1985 auf US$ 573 Mrd. im Jahre 2003, mit einem zwischenzeitlichen Höchststand von US$ 1511 Mrd. im Jahr 2000. Diese Entwicklung lässt sich auch als Anteil der Auslandsdirektinvestitionen am Bruttoinlandsprodukt weltweit darstellen (siehe Abb. 1). Die Tatsache führte dazu, dass Ökonomen ein zunehmendes Interesse an den Ursachen und Folgen dieser Entwicklungen fanden und immer stärker nach Gründen suchten, die für die zunehmende multinationale Ausrichtung von Unternehmen Erklärungen abgaben. Im Folgenden werden zwei Theorieansätze sowie ein empirischer Teil dargestellt, die versuchen, den internationalen Handel durch multinationale Unternehmen zu erklären. D.h. sie geben Antworten auf die Frage, warum Länder miteinander Handel treiben, welche Güter exportiert und welche importiert werden und welche Anpassungsprozesse bei der Existenz von Handelsbarrieren ablaufen. Das Ziel dieser Arbeit ist es weiterhin die Gründe, sowie die unterschiedlichen Möglichkeiten eines Unternehmens sich multinational auszurichten, darzustellen, und zu zeigen, welche Bedingungen bzw. Vorraussetzungen hierfür gegeben sein müssen. Ein Unternehmen muss verschiedene Entscheidungen treffen, bevor es eine Investition im Ausland tätigt. Falls man sich jedoch entschieden hat zu investieren bleibt immer noch die Frage, in welcher Form und in welchem Ausmaß man das tun sollte: den kompletten Produktionsprozess ins Ausland verlagern (horizontale Integration) oder die Produktion von Zwischen- und Endprodukten international auf verschiedene Länder aufteilen (vertikale Integration)? Im Fokus der Arbeit steht die Beantwortung dieser Frage, die Vor- und Nachteile der beiden Formen für die Unternehmung, unter welchen Vorraussetzungen solche Unternehmen entstehen, welche Handelsmuster sich daraus ableiten lassen sowie die Auswirkungen auf die Ziel- und Herkunftsländer der Investitionen. Gang der Untersuchung: In Kapitel 2 werden wir uns zunächst mit den Begriffen der Auslandsdirektinvestition und der multinationalen Unternehmung auseinandersetzen und diese definieren. Weiterhin werden spezielle Formen der Auslandsdirektinvestition, namentlich die vertikale und die horizontale Auslandsdirektinvestition, vorgestellt und die jeweils wesentlichen Erkennungsmerkmale dieser genannt. Kapitel 2 schließt mit einigen Zahlen und Fakten, die die Bedeutung von Auslandsdirektinvestitionen in unserer globalisierten Welt unterstreichen und stellt neueste Trends und Entwicklungen dar. In Kapitel 3 beginnt der eigentliche Hauptteil. Er beinhaltet das erste theoretische Modell dieser Arbeit und beschäftigt sich mit der horizontalen Integration einer multinationalen Unternehmung. Zunächst wird das Grundmodell vorgestellt, welches im folgenden Teil in der Komparativen Statik untersucht wird. Nach den Ergebnissen der Komparativen Statik folgt eine grafische Darstellung des Modells. Schließlich endet Kapitel 3 mit einer kritischen Betrachtung des Modells. In Kapitel 4 wird das zweite theoretische Modell dieser Arbeit dargestellt. Dieses beschäftigt sich mit der vertikalen Integration der multinationalen Unternehmung. Es beginnt ebenfalls mit der Vorstellung des Grundmodells, fährt mit einer grafischen Analyse fort, geht anschließend auf multinationale Aktivitäten innerhalb des Modells ein und endet schließlich wiederum mit einer kritischen Betrachtung des Modells. In Kapitel 5 wird das theoretische Hybrid aus horizontaler und vertikaler Auslandsdirektinvestition, das sogenannte Knowledge-Capital-Modell, vorgestellt und anschließend auf seine Konsistenz mit der Realität empirisch getestet. Horizontale Auslandsdirektinvestitionen dienen dazu, die mit dem Export der Produkte verbundenen Kosten, durch Gründung eines Tochterunternehmens bzw. Beteiligung an einem Unternehmen, zu umgehen. Dabei wird das Tochterunternehmen bzw. das beteiligte Unternehmen als Exportplattform für den ausländischen Mutterkonzern eingesetzt. Durch diese Form können, wie bereits genannt, typische Marktzutrittsbarrieren (Zölle, etc.) umgangen werden. Die Unternehmung kopiert ihre Aktivitäten aus dem Heimatland und überträgt diese auf die ausländischen Produktionsanlagen. Bei vertikalen Auslandsdirektinvestitionen wird der Produktionsprozess geografisch aufgeteilt, was bedeutet, dass die verschiedenen Produktionsstufen teilweise oder vollständig ins Ausland ausgelagert werden. Die Grundidee ist, dass der Produktionsprozess aus verschiedenen Stufen besteht, die jeweils unterschiedliche Vorleistungen (z.B. Arbeit, Kapital, etc.) benötigen. Wenn die Preise für diese zwischen den Ländern variieren, kann es profitabel für die Unternehmung sein, die Produktionsstufen aufzuteilen. Vertikale Auslandsdirektinvestitionen können entweder vorwärts oder rückwärts gerichtet sein. Im Falle der rückwärts gerichteten ADI richtet sich die multinationale Unternehmung ihren eigenen Zulieferer ein, der sie mit Vorleistungen bzw. benötigten Inputs beliefert. Im Falle der vorwärts gerichteten ADI gründet das Mutterunternehmen eine ausländische Tochtergesellschaft, die ihrerseits dann Vorleistungen zur weiteren Produktion vom Mutterunternehmen erhält und an nachgelagerter Stelle im Produktionsprozess angesiedelt ist. Die Bedeutung von Auslandsdirektinvestitionen und neuere Trends: In den letzten zwanzig Jahren hat sich das Investitionsklima in den verschiedenen Ökonomien der Welt durchaus positiv entwickelt. Es wurde begünstigt durch die vorangetriebene Deregulierung und auch Privatisierung in vielen Staaten. Auslandsdirektinvestitionen sind ein fester Bestandteil der Unternehmensstrategie geworden, was wiederum zu diesem enormen Anstieg von grenzüberschreitenden Investitionen führte. Wie wir bereits in Abbildung 1 gesehen haben, wuchs der Anteil der Auslandsdirektinvestitionen am Bruttoinlandsprodukt in den vergangenen Jahrzehnten stetig an. Während der Anteil noch in den Siebziger Jahren bei knapp 0,5% lag, so stieg er im Jahre 2000 auf 6% an, fiel danach jedoch wieder auf den heutigen Stand von ca. 2% ab. Das Unternehmensklima erholte sich in den Jahren 2004 und 2005 von der Rezession in den drei Jahren zuvor und erreichte 2005 den vierthöchsten Stand an Zuflüssen (in OECD-Länder) seit Aufzeichnung der Daten und hatte einen Wert von US$ 622 Milliarden. Innerhalb der OECD-Länder war Großbritannien der attraktivste Standort für Investitionen. Außerhalb der OECD-Länder ging die wirtschaftliche Entwicklung noch schneller voran, wobei hier China, gefolgt von Indien, als attraktivste Standorte für Direktinvestitionen zu nennen sind. Betrachtet man die geografische Verteilung von ADI etwas genauer, so stellt man fest, dass der Großteil an ADI immer noch zwischen den entwickelten Ländern stattfindet - wobei die Entwicklungsländer deutlich auf dem Vormarsch sind (siehe Abbildung 2: Zu- und Abflüsse von Auslandsdirektinvestitionen nach Regionen aufgeteilt). Wie aus Abbildung 2 hervorgeht, waren die Europäische Union zusammen mit den USA noch im Jahre 2000 für mehr als 70% der Zuflüsse von ADI und mehr als 80% der Abflüsse von ADI verantwortlich. Sowohl die Zuflüsse als auch die Abflüsse an ADI waren im Jahre 2000 mehr als 6mal so hoch als noch zehn Jahre zuvor (in absoluten Werten gemessen). Diese Tatsache kann man auch an der Wachstumsrate der Auslandsdirektinvestitionen in Abbildung 5 sehen. Parallel zur Entwicklung der Auslandsdirektinvestitionen verlief der Anstieg von multinational aktiven Unternehmen. Dies ist jedoch nicht verwunderlich, da diese ja die treibenden Kräfte in diesem Zusammenhang darstellen. So gab es laut UNCTAD Ende der Sechziger Jahre nur rund 7000 multinationale Unternehmen und Mitte der Neunziger schon rund 40.000. Die Theorie der horizontalen Integration: Im ersten Hauptteil dieser Arbeit wird ein Modell der horizontalen Integration der multinationalen Unternehmung vorgestellt und analysiert. Unter welchen Annahmen und welchen Vorraussetzungen haben diese Unternehmen einen Anreiz multinational aktiv zu werden? Sollen sie den ausländischen Markt über Exporte versorgen oder eigene Produktionsstätten im Ausland errichten? Errichtet das Unternehmen eine eigene Produktionsanlage im Ausland, so sieht es sich mit verschiedenen zusätzlichen Kosten konfrontiert. Darunter fallen z.B. die Kosten sich mit ausländischen Regierungen, Rechtsvorschriften und anderen Steuersystemen auseinanderzusetzen. Auf der anderen Seite kann das Unternehmen durch eine lokale Produktion im Ausland auch einige Kosten einsparen. Die offensichtlichsten Kosten hierbei sind die Transportkosten oder Zölle, die im Falle einer horizontalen Integration eingespart werden können. Die in der Literatur bekannten theoretischen Modelle zur horizontalen Integration beinhalten genau diesen typischen Trade-Off der Unternehmung. Welche Kosten sind geringer - die zusätzlichen Fixkosten, die durch die Errichtung einer neuen Produktionsanlage entstehen oder die variablen Kosten (Transportkosten, etc.), die beim Exportieren der Produkte entstehen? Diese und weitere Fragen werden im nun folgenden Modell erörtert und dargestellt. Das Ziel dieses Modells ist es, ein flexibles Konstrukt zu bilden, welches nationalen Unternehmen (im weiteren Verlauf der Ausführungen "NE" genannt) und multinationalen Unternehmen (im Folgenden "MNE" genannt) erlaubt, je nach zu Grunde liegenden Parametern im Markt endogen zu entstehen. Das Modell ist so gewählt, dass es mit den Standard-Modellen der neuen Handelstheorie verglichen werden kann. Wir konzentrieren uns in diesem Modell auf horizontale Auslandsdirektinvestitionen, in denen multinationale Unternehmen dasselbe Produkt in mehreren Produktionsstätten herstellen. Es gibt bereits mehrere vorangehende Modelle. So enthalten diese auch die Unterscheidung zwischen firmenspezifischen und produktionsanlagespezifischen Skaleneffekten und das Vorhandensein von Transportkosten und Zöllen. In unserem Modell versuchen wir zu analysieren, warum horizontale Auslandsdirektinvestitionen eher zwischen Ländern entstehen, die bezüglich ihrer Größe, Ausstattung und Technologie ähnlich sind. Gegenstand der Untersuchung ist die Umsetzung der europäischen, gleichstellungspolitischen Strategie Gender Mainstreaming (GM) in der Ministerialverwaltung des neuen EU-Mitgliedslands Estland. GM hat die Umsetzung der Gleichstellung von Männern und Frauen zum Ziel und wird als eine Querschnittsaufgabe mit Instrumenten der Verwaltungsmodernisierung (Folgenabschätzung, Wissensmanagement, u.a.) umgesetzt. Wie diese Strategie in der Ministerialverwaltung als ausführendes Organ der Regierung aufgenommen, übersetzt und umgesetzt wird in einem Land, das viele Jahrzehnte dem kommunistischen Gleichheitspostulat unterworfen war und als Staatsneugründung seine nationale Verwaltung erst aufbauen musste, wird in der Arbeit beschrieben und analysiert. Die Dissertation ist in vier Teile gegliedert: in Teil I wird in den Untersuchungsgegenstand und die Methode der Arbeit eingeführt. Teil II beschreibt die gesellschaftlich-politischen und administrativen Rahmenbedingungen im Fallbeispiel Estland. Teil III widmet sich dem Untersuchungsgegenstand "Umsetzung von GM in der estnischen Ministerialverwaltung". Der IV. Teil beschließt die Arbeit mit der Analyse der Zusammenhänge zwischen den Rahmenbedingungen und der Umsetzung. Teil I beginnt mit der Darstellung des Forschungskonzepts, das sich aus Elementen der Verwaltungswissenschaft und der Forschung zu staatlichen Strukturen für Gleichstellungspolitik, einem Zweig der politikwissenschaftlichen Geschlechterforschung, zusammensetzt. Damit wird für die Untersuchung von GM erstmals systematisch die Verwaltungswissenschaft herangezogen. Die Arbeit wird methodisch und theoretisch als explorativ-explanative Single Case Studie verortet, die sich an neo-institutionalistischen Ansätzen orientiert. Teil II der Arbeit führt in das Fallbeispiel Estland ein: Es werden drei identifizierte Interpretationsmuster dargestellt anhand derer in Estland die Vergangenheit als besetzte Nation und die Gegenwart als demokratischer Staat (re )konstruiert werden und die das estnische, kollektive Selbstverständnis prägen. Anschließend werden die gesellschaftlichen und administrativen Rahmenbedingungen und Einflussfaktoren beschrieben, die für die Umsetzung von Querschnittsreformen in der öffentlichen Verwaltung und für die Umsetzung von Gleichstellungspolitik von Bedeutung sind. Die Forschungsergebnisse in Teil II zeigen über die empirischen Befunde hinaus, dass Estland nicht immer eindeutig in klassische politikwissenschaftliche Kategorien einzuordnen ist. Sowohl die Transitionssituation des Landes als auch die an westlichen Demokratien ausgerichteten Untersuchungskriterien sind für diesen Befund ursächlich. Teil III der Arbeit widmet sich dem Untersuchungsgegenstand GM. Nach grundlegenden Informationen zu dieser Verwaltungsmodernisierungsstrategie folgt die Darstellung der Umsetzung in der estnischen Ministerialverwaltung. In Teil IV der Dissertation werden die in Teil II beschriebenen Variablen auf die Umsetzung von GM (Teil III) bezogen. Die Analyse erfolgt anhand von Kriterien, die sich aus der Auswertung internationaler GM-Implementierungserfahrungen ergeben. Die Untersuchung zeigt, dass das post-kommunistisch geprägte, gesellschaftliche Klima besondere Legitimitätsprobleme für eine an Gleichheit orientierte staatliche Politik schafft. Dies kann die schwache zivilgesellschaftliche gleichstellungspolitische Lobby nur sehr begrenzt beeinflussen. Die strukturellen Bedingungen der estnischen Ministerialverwaltung mit ihrer geringen Koordinationsfähigkeit und politischen Steuerbarkeit machen eine effektive Umsetzung von Querschnittsreformen allgemein schwierig. Als produktiv für die Umsetzung hat sich der hohe Grad der fachlichen Professionalität und Politikversiertheit der kleinen, gleichstellungspolitischen Elite in der Ministerialverwaltung herauskristallisiert. Über Kooperationen mit internationalen Akteuren und estnischen zivilgesellschaftlichen Kräften sowie einzelnen interessierten Personen in der Verwaltung treibt sie die Umsetzung von GM voran. Sie nutzte die EU-Beitrittsverhandlungen um politischen Handlungsdruck für die Verwaltungsmodernisierung durch GM aufzubauen. Nachdem dieser seit dem Beitritt nicht aufrecht erhalten werden kann, zeichnet sich eine neue Umsetzungsstrategie ab. Es wird zukünftig nicht mehr vor allem an den normativen und kognitiven Strukturen in der Verwaltung, also den Einstellungen und Fachkompetenzen des Verwaltungspersonals zu gleichstellungsorientierter Arbeit, angesetzt. Vielmehr sollen neue, gleichstellungsrelevante Wissensbestände durch Expertinnen und Experten und exponierte Persönlichkeiten in die Gesellschaft und die Verwaltung transportiert und damit grundlegende gesellschaftlich-normative Voraussetzungen für die Rezeptivität von GM verbessert werden. ; The object of research is the implementation of the European equal opportunities policy 'gender mainstreaming' (GM), in the ministerial administration of the new European Union member state Estonia. GM is a transversal task that employs the instruments of administration modernisation (impact assessment, knowledge management and others), its aim is equal opportunities for men and women at all levels. This paper describes and analyses how GM strategy is received, interpreted and implemented by the ministerial administration, as executive body of government, in a country that has had to rebuild its national administration following years of subjugation to the communist egality postulate. The dissertation is divided into four parts. Part I introduces the research object and methodology. Part II describes the socio-political and administrative framework of the case example Estonia. Part III is devoted to the research object 'Implementation of GM in the Estonian ministerial administration'. Part IV concludes the paper with an analysis of the correlations between the socio-political parameters and the implementation. Part I begins with the presentation of the research concept that comprises elements from Public Administration and research on state structures for equal opportunities policy, a field of gender studies in political science. This means that for the first time the science of public administration is being systematically applied to GM research. The research employs explorative/explanative single case study methods and theory in accordance with neo-institutionalism approaches. Part II introduces the case study Estonia describing three identified interpretation models on the basis of which Estonia is (re)-constructing its past as an occupied nation and the present day as a democratic state, and which characterise Estonian national identity. Finally, an account is given of the social and administrative parameters and the influencing factors significant for the implementation of both transversal reforms in public administration and equal opportunities policies. Research results in part II go beyond the empirical findings and show that Estonia cannot always be conclusively classified within standard political science categories. This is due both to the country's transitional situation as well as to research criteria that are tailored to Western democracies. Part III is devoted to the research object GM. Key information on this modernisation strategy is followed by a report on its implementation in the Estonian ministerial administration. In part IV of the dissertation the variables described in part II are related to the implementation of GM (part III). This analysis is carried out on the basis of criteria resultant from the evaluation of international experience of GM implementation. The research shows that the social climate of the post-communist era generates specific legitimacy problems for a national policy based on equality, upon which the powerless equal opportunities lobby in civil society has only limited influence. The structural conditions in the Estonian ministerial administration, with their limited coordinating ability and political governance, render effective implementation of transversal reforms generally quite difficult. It has emerged that the small equal opportunities 'elite' in the ministerial administration with their high degree of professionalism and political experience is /the /crucial factor for the GM implementation process. Through cooperation with international players and powers within Estonian civil society, as well as interested individuals in the public administration, they are able to keep implementation of GM going. They used EU-membership negotiations to build political pressure to modernise the public administration with GM. Following EU-membership this could not be sustained and a new implementation strategy is now beginning to emerge. In future, efforts will no longer be concentrated mainly on the normative and cognitive structures in the administration, meaning the attitudes and professional competences of the administrative staff with regard to equal opportunities. Instead, the intention is to transport new corpuses of knowledge relevant to equal opportunities into society as a whole, and public administration in particular, by experts and persons in exposed positions. In this way the elite expects fundamental socio-normative preconditions for receptivity of GM to be influenced for the better.

Mit der Verabschiedung der Flora-Fauna-Habitat-Richtlinie (FFH-RL) im Jahr 1992 wurde ein Meilenstein für den Naturschutz auf europäischer Ebene gesetzt. Alle EU-Mitgliedsstaaten sind demnach verpflichtet, ein europaweit zusammenhängendes Netz von Schutzgebieten (nach FFH-RL und nach Vogelschutz-Richtlinie) unter dem Namen Natura 2000 zu schaffen. In der FFH-RL wird ausdrücklich die Überwachung von Lebensraumtypen und Arten gefordert. Es sind umfassende Daten zu ihrem Vorkommen, zur Verbreitung und zu ihrem Erhaltungszustand zu sammeln. Alle sechs Jahre ist ein ausführlicher Bericht darüber abzulegen, so dass mit der FFH-Richtlinie die erste gesetzlich verankerte Pflicht zum flächenhaften Monitoring in der EU eingeführt wurde. Die Erfassung von aktuellen Daten zur Erfüllung dieser Monitoringaufgaben ist mit einem beträchtlichen Zeit- und Kostenaufwand verbunden. Es besteht die Notwendigkeit, geeignete Konzepte und Methoden für die Datenerhebung zu entwickeln. Im Hinblick auf den hohen und wiederkehrenden Informationsbedarf für die Erfüllung der FFH-Berichtspflichten ist der Einsatz der Fernerkundung unbedingt notwendig. Die traditionell verwendeten analogen Luftbilder sind in ihrer Herstellung und Auswertung für die finanziell schlecht ausgestatteten Verwaltungen nicht mehr zu bezahlen. Die neue Generation höchstauflösender Satelliten (v.a. Quickbird) bietet die Möglichkeit, kleine und weit verstreute Gebiete kostengünstig zu überwachen. Es fehlen jedoch geeignete automatisierte Methoden für die Auswertung der Satellitenbilder, die im Sinne der FFH-Richtlinie einsetzbare Ergebnisse liefern könnten. Die wiederholte Bewertung des Erhaltungszustandes von Lebensraumtypen kann nur auf der Grundlage von Indikatoren objektiv und übertragbar erfolgen. Biotopelemente (z.B. der Gehölzanteil) sind sehr wichtige Indikatoren für die Bewertung der Habitatstruktur und von eventuell vorhandenen Beeinträchtigungen. Vor diesem Hintergrund war das Ziel dieser Arbeit, ein Verfahren zu entwickeln, das höchstauflösende Satellitendaten für die Erfassung von Indikatoren für das Monitoring ausgewählter FFH-Lebensraumtypen der Offenbereiche einsetzt und das dabei eine möglichst weitgehende Automatisierung erreicht. Es wurde ein wissensbasierter Ansatz gewählt, der mit Hilfe allgemeingültiger Regeln und unter Einbeziehung von a-priori Information (z.B. Biotoptypen- und Landnutzungskartierung) eine vollautomatische Klassifikation von Biotopelementen ermöglicht. Das Verfahren arbeitet auf Pixelbasis und stellt eine hybrides hierarchisches Klassifikationssystem dar. Die Flächengrenzen der Lebensraumtypen und die Genauigkeit der Klassifikation müssen im Anschluss visuell überprüft werden, so dass es sich insgesamt um eine semi-automatische Vorgehensweise handelt. Das Verfahren wurde anhand vier ausgewählter Untersuchungsgebiete in Brandenburg für zwölf unterschiedliche Lebensraumtypen der Offenbereiche entwickelt und validiert. Die Übertragbarkeit auf andere Gebiete, andere Grundlagendaten und andere Auflösungsstufen wurde ausführlich in vier weiteren Untersuchungsgebieten (in Brandenburg, Thüringen und Schleswig-Holstein) getestet. Für die untersuchten Lebensraumtypen der Halbtrockenrasen und Moore gab es aufgrund der Ausstattung der Testgebiete nur sehr wenig Vorkommen und terrestrisch verifizierte Grundlagendaten, diese sollten weiter untersucht werden. Für die Ermittlung der Indikatoren für Heidehabitate, für Dünen mit offenen Sandtrockenrasen und für Süßwasserlebensräume ist das Verfahren an sehr vielen Flächen erfolgreich getestet worden und kann operationell eingesetzt werden. Der Einsatz der höchstauflösenden Satellitenbilder und des Klassifikationsverfahrens kann das Monitoring von FFH-Lebensraumtypen insgesamt erheblich erleichtern. Es ist möglich, für die untersuchten Lebensraumtypen Strukturindikatoren und Beeinträchtigungen abzuleiten sowie die Flächengrenzen zu überprüfen. Dazu kommt die Erstellung aktueller Biotoptypen- und Landnutzungsdaten und die Verfügbarkeit aktueller digitaler Orthofotos für die terrestrische Kartierung. Große Flächen können schnell bearbeitet werden und die Trefferquote und Konstanz der visuellen Interpretation kann mit der automatischen Klassifikation wesentlich erhöht werden. Die Flächenanteile wichtiger Indikatoren sind sehr genau berechenbar und eine gute Grundlage nicht nur für das Monitoring, sondern auch für die detaillierte Maßnahmenplanung. ; The adoption of the EC Habitats Directive (FFH-RL) in 1992 was an important milestone for nature conservation on the European level. The provisions of the Directive require every Member State to establish a coherent net of Special Areas of Conservation (SAC) embraced with the Special Areas of Protection (SPA) from the EC Birds Directive under the name Natura 2000. Furthermore the EC Habitats Directive claims the introduction of a range of different measures like the surveillance of habitats and species, a report on species conservation and a report on the implementation of the Directive. An extensive amount of data on occurrence, geographical range, representativity and conservation status of habitats and species is required. Every six years a report on the implementation of the Directive is due, which incorporates the first statutory obligation on area-wide surveillance within the European Union. The collection of actual information with regard to these obligations is associated with considerable amounts of time and expenses. Thus applicable concepts and methods for data acquisition have to be developed. Facing the high and repeated information demand the employment of remote sensing is necessary. Traditionally analogue air photos were used for vegetation monitoring, but their acquisition and interpretation are no longer affordable for the Nature Conservation Administration in Germany. The new generation of digital very high resolution satellites (particularly Quickbird) offers the opportunity for a cost-efficient survey of small and widespread areas. But suitable automated methods for the analysis of those satellite data in terms of the EC Habitats Directive are missing. The repeated and impartial assessment of the conservation status of habitat types can only be based on indicators. Elements of biotopes (e.g. percentage of shrubs) are very useful indicators for the assessment of habitat structure and impairments. The aim of this study was to develop a method for the analysis of very high resolution satellite data, that can be used for the acquisition of indicators to support the monitoring of a selected subset of non-forested habitats and that operates to a large extent in an automated mode. With a knowledge-based procedure, implementing universal rules and integrating a-priori information (e.g. biotope type and land use maps), a fully-automated way was found to classify biotope elements. The procedure is pixelbased and works with a hybrid and hierarchical classification scheme. Subsequent to the classification the habitat borders and the classification accuracy have to be evaluated visually. The method was developed and validated for 12 different non-forested habitat types on the basis of four test sites in Brandenburg. The transferability on other sites, other a-priori information and different geometric resolutions was tested in detail with the help of four additional test sites (in Brandenburg, Thuringia and Schleswig-Holstein). For the investigated habitat types that represent bogs and dry grasslands only few habitat areas were available due to the character of the test sites, they should be examined further. For the investigated habitat types that represent dry heaths and inland dunes and for freshwater habitat types the procedure has been successfully validated on the basis of many areas and can be used operationally. The use of very high resolution satellite data and the developed classification procedure can substantially support the monitoring of habitat types. It was possible to automatically derive indicators assessing the structure and existing impairments of the investigated habitat types. The habitat borders have to be verified visually. Additional results of the procedure are actual biotope type and land use maps and digital ortho photos as an important support for the terrestrial mapping. Large areas can be processed in short time and also the accuracy and stability of the visual interpretation increase with help of the automated classification. Percentages of important indicators can be calculated on a high level of detail. They represent a very good basis not only for the monitoring but also for the explicit planning of conservation measures.

Rychlik, Czech Republic. ; The aims of this study were to determine the frequency of dialysis and kidney transplantation and to estimate the regularity of comprehensive conservative management (CCM) for patients with kidney failure in Europe. This study uses data from the ERA-EDTA Registry. Additionally, our study included supplemental data from Armenia, Germany, Hungary, Ireland, Kosovo, Luxembourg, Malta, Moldova, Montenegro, Slovenia and additional data from Israel, Italy, Slovakia using other information sources. Through an online survey, responding nephrologists estimated the frequency of CCM (i.e. planned holistic care instead of kidney replacement therapy) in 33 countries. In 2016, the overall incidence of replacement therapy for kidney failure was 132 per million population (pmp), varying from 29 (Ukraine) to 251 pmp (Greece). On 31 December 2016, the overall prevalence of kidney replacement therapy was 985 pmp, ranging from 188 (Ukraine) to 1906 pmp (Portugal). The prevalence of peritoneal dialysis (114 pmp) and home hemodialysis (28 pmp) was highest in Cyprus and Denmark respectively. The kidney transplantation rate was nearly zero in some countries and highest in Spain (64 pmp). In 28 countries with five or more responding nephrologists, the median percentage of candidates for kidney replacement therapy who were offered CCM in 2018 varied between none (Slovakia and Slovenia) and 20% (Finland) whereas the median prevalence of CCM varied between none (Slovenia) and 15% (Hungary). Thus, the substantial differences across Europe in the frequency of kidney replacement therapy and CCM indicate the need for improvement in access to various treatment options for patients with kidney failure. ; Peer reviewed

Σε αυτή την σειρά άρθρων προσπαθούμε να προσδιορίσουμε τους προσδιοριστικούς παράγοντες των εισερχόμενων άμεσων ξένων επενδύσεων (ΑΞΕ) δίνοντας ιδιαίτερη έμφαση στο ανθρώπινο κεφάλαιο και την εκπαίδευση κατά φύλο. Η παρούσα διδακτορική διατριβή αποτελείται από τρεις ανεξάρτητες εργασίες, εκ των οποίων η πρώτη είναι αφιερωμένη αποκλειστικά στην μελέτη του ανθρωπίνου κεφαλαίου, η δεύτερη στην επίδραση της εκπαίδευσης με βάση το φύλο λαμβάνοντας υπόψιν διαφορετικά είδη και επίπεδα εκπαίδευσης, και η τρίτη στον τρόπο με τον οποίο η εκπαίδευση των γυναικών και η κουλτούρα διαμορφώνει τις εισερχόμενες ΑΞΕ λαμβάνοντας υπόψιν το οικονομικό, εκπαιδευτικό και θεσμικό περιβάλλον. Η πρώτη εργασία, που παρουσιάζεται στο Κεφάλαιο 2, αξιολογεί το ρόλο του ανθρωπίνου κεφαλαίου στην προσέλκυση ΑΞΕ στις χώρες της Ευρωπαϊκής Ένωσης (ΕΕ). Ειδικότερα, παρέχει μια αναλυτική διερεύνηση του ρόλου του ανθρωπίνου κεφαλαίου, των δεξιοτήτων και των ικανοτήτων στην εγκατάσταση των ΑΞΕ συγκρίνοντας τις Δυτικές (ΕΕ-15) και τις Κεντρικές και Ανατολικές (ΕΕ-11) χώρες της ΕΕ. Σε αντίθεση με τις προϋπάρχουσες μελέτες εξετάζουμε μία εκτεταμένη λίστα παραδοσιακών και καινούριων προηγμένων μέτρων που συλλαμβάνουν το ανθρώπινο κεφάλαιο, ακόμη και διαφορετικών σχολικών συστημάτων. Αξιολογούμε, για πρώτη φορά, στην ιστορία των ΑΞΕ, την τεχνολογική έναντι της γενικής εκπαίδευσης, ακολουθώντας τα Τεχνολογικά και Εκπαιδευτικά Προγράμματα της ΕΕ καθώς και την ποιότητα του ανθρωπίνου κεφαλαίου όπως μετράται με διεθνείς βαθμολογίες. Τα αποτελέσματα υποδεικνύουν μια κύρια διαφορά σχετικά με τα θεωρητικά και τεχνολογικά προγράμματα εκπαίδευσης για τις δύο υπο-περιφέρειες – επιπλέον, λαμβάνουμε μία κύρια διαφορά για συγκεκριμένα προσόντα στις διεθνείς βαθμολογίες. Εξετάζουμε διάφορα μέτρα ανθρωπίνου κεφαλαίου με βάση το φύλο, ώστε να συλλάβουμε την περίπλοκη φύση του ανθρωπίνου κεφαλαίου και της εκπαίδευσης αναφορικά με το επίπεδο και το είδος της εκπαίδευσης και να δούμε τι είναι πιο σημαντικό στα μάτια των ξένων επενδυτών. Χρησιμοποιώντας εκτιμήσεις διαστρωματικών δεδομένων, συμπεραίνουμε ότι η μείωση των εκπαιδευτικών ανισοτήτων με βάση το φύλο και στις δύο υπο-περιφέρειες διευκολύνει την απορρόφηση των εισερχόμενων ΑΞΕ. Οι υπεύθυνοι χάραξης πολιτικής θα πρέπει να ενισχύσουν την ισότητα με βάση το φύλο στην τεχνολογική εκπαίδευση στα παλαιότερα μέλη της ΕΕ ενώ προσοχή θα πρέπει να δοθεί στην ισότητα στην γενική εκπαίδευση στις Κεντρικές και Ανατολικές χώρες της ΕΕ. Η τρίτη εργασία (Κεφάλαιο 4) εξετάζει τον τρόπο με τον οποίο η κουλτούρα, η θρησκεία και η εκπαίδευση διαμορφώνουν τις ΑΞΕ στις χώρες της Ευρωπαϊκής Ένωσης για την περίοδο 2000-2012. Αναφορικά με την εκπαίδευση, επικεντρωνόμαστε στην τριτοβάθμια εκπαίδευση και στους τομείς Φυσικής, Τεχνολογίας, Μηχανικής και Μαθηματικών (γνωστών ως STEM – Science, Technology, Engineering and Mathematics) – οι τομείς STEM είναι ουσιαστικής σημασίας για τη σύγχρονη αγορά εργασίας. Ωστόσο, το ποσοστό αποφοίτησης στην ΕΕ είναι πολύ χαμηλό. Τα αποτελέσματα αποκαλύπτουν ότι όχι μόνο οι απόφοιτοι της τριτοβάθμιας εκπαίδευσης αλλά και οι απόφοιτοι της τριτοβάθμιας εκπαίδευσης σε τομείς STEM είναι σημαντικοί παράγοντες για την προσέλκυση ΑΞΕ. Το Κεφάλαιο 5 απεικονίζει τα συμπεράσματα και τονίζει ορισμένες πολιτικές κατευθύνσεις σχετικά με το πώς το ανθρώπινο κεφάλαιο, η εκπαίδευση κατά φύλο και ιδιαίτερα η εκπαίδευση των γυναικών τονώνουν τις ΑΞΕ και πού θα πρέπει να επικεντρώνονται περισσότερο οι υπεύθυνοι χάραξης πολιτικής. ; This series of papers tries to reflect the determinants of inward FDI by placing particular emphasis on human capital, gender disparities in terms of education and even more so on the role of females' education in science fields. The present dissertation consists of three independent essays, of which the first one is devoted completely to the study of human capital, skills and competencies, the second to gender disparities effects in terms of different types and levels of education, and the third to how females' education and culture shape the FDI inflows taking also into account the economic, human capital and institutional environment. The first essay, which is presented in Chapter 2, evaluates the role of human capital in attracting inward foreign direct investment (FDI) in EU countries. Specifically, it provides a comprehensive investigation on the role of human capital, skills and competencies in the location of inward FDI by comparing Western (EU15) and Central and Eastern (CEE) European Union (EU) members. We go beyond existing studies by examining an extensive list of traditional and newly advanced measures capturing human capital, even differing schooling systems as well as skills and competencies. We assess, for the first time in the FDI literature, vocational vs. general education, following the Vocational and Educational Programs of the EU as well as quality human capital aspects as captured by international scores. Results indicate a major difference regarding theoretical and vocational education programs for both sub-regions; in addition, a major difference is obtained for particular qualifications in international scores. There is evidence for potential internal and external inefficiency in education in CEE countries in contrast to the EU15, which calls for re-evaluation and restructuring in their education system towards more efficient use of funds; this would earn investors' trust and meet labor markets' demands, thus stimulate more and higher value added foreign investments. Results also point to useful implications for managers who should watch closely education reforms. The second essay, presented in Chapter 3, is a systematic analysis of the relationship between gender educational disparities and inward FDI in Western and CEE EU countries to detect potential differentiations between them. We examine various gender related human capital measures so as to capture the complicated nature of human capital and education concerning level and type with respect to gender to see which is more important in the eyes of foreign investors. Using panel data estimations, we conclude that the reduction of gender educational gaps in both sub-regions facilitates the absorption of inward FDI. Policymakers should enhance gender equality in vocational education in Western EU members while attention should be given in gender equality in theoretically oriented programmes in the CEE EU ones. The third essay (Chapter 4) examines how national culture, religion, and females' education, parts of institutions, shape the FDI inflows in a panel of European Union countries for the period 2000-2012. In terms of education, we focus on tertiary level and Science, Technology, Engineering and Mathematics (STEM) fields; the latter is vital in the modern labor market. However, the females' graduate rate in EU is very low. The results reveal that not only tertiary education graduates but even more so tertiary graduates with STEM skills are important in attracting FDI. Moreover, the presence of high female STEM labor force is particularly an attracting feature for FDI. Finally, institutions, culture and the economic environment are important determinants in attracting FDI. Finally, Chapter 5, presents the concluding remarks of all essays and highlights some policy implications of how human capital, gender education and particularly females' education stimulate FDI and where should policymakers pay most attention.

- Objetivos. 1. Caracterización de la longitud telomérica y de la expresión y actividad de la telomerasa en el pez cebra (Danio rerio) a lo largo de su ciclo de vida. 2. Caracterización de la línea de pez cebra deficiente en la subunidad catalítica de la telomerasa (tert). 3. Estudio de la longitud telomérica y de la expresión y actividad de la telomerasa en el pez cebra (Danio rerio) durante el proceso de regeneración de la aleta caudal del pez cebra salvaje y del pez cebra tert-deficiente. - Metodología. Para el primer objetivo, se han utilizado peces cebra de tipo salvaje, de diferentes estirpes y edades. El estudio de la expresión de genes se ha llevado a cabo mediante RT-PCR y PCR a tiempo real, el estudio de la actividad telomerasa mediante una técnica específica y cuantitativa llamada Q-TRAP basada en la amplificación de la secuencia telomérica. La longitud telomérica se ha cuantificado por técnicas diferentes: TRF que consiste en una hibridación con una sonda telomérica radioactiva, Q-FISH y Flow-FISH que consisten en una hibridación in situ con una sonda telomérica fluorescente. Para el cumplimiento del segundo objetivo, se han utilizado los genotipos, tert+/+, tert+/- y tert-/-. Además de las técnicas antes citadas, se han realizado curvas de supervivencia y un estudio histológico sobre diferentes tejidos (Hematoxilina/Eosina, PAS y TUNEL). El estudio de la G2 mutante se ha llevado a cabo con curvas de supervivencia, ensayo de apoptosis por TUNEL, y Q-FISH en núcleos metafásicos. Para el tercer objetivo, con los mismos genotipos y a diferentes edades. Se ha realizado un ensayo de detección de los círculos de ADN telomérico, un ensayo de regeneración en adultos con inhibición de un gen utilizando un morfolino seguido de electroporación o en larvas con inhibición de un gen por inmersión. Peces cebra han sido mantenidos según se indica en el manual del pez cebra. Todos los experimentos cumplen con la directiva de la Unión Europea (86/609/EU) y han sido aprobados por el Comité de Bioética del Hospital Clínico Universitario "Virgen de la Arrixaca" (España) y por el Comité Institucional del Cuidado y Uso Animal del Hospital Infantil de Boston (EEUU). - Resultados y conclusiones. Los resultados de esta tesis han demostrado la estrecha relación de la expresión de la telomerasa y la longitud de los telómeros durante la vida del pez, concluyendo que estos dos parámetros pueden ser utilizados como biomarcadores de envejecimiento en el pez cebra. La línea de pez cebra deficiente en telomerasa ha mostrado signos de envejecimiento prematuro, como curvatura de la columna, infertilidad y reducción de la vida media de la G1, apoyando al pez cebra como un modelo prometedor para estudiar el envejecimiento inducido por el telómero. Además, la G1 muestra activación de p53 en respuesta al desgaste telomerico, reproduce el fenómeno de anticipación genética, ligado al acortamiento telomérico observado en humanos con DC, debido a haploinsuficiencia de la telomerasa. La G2 muere en las primeras etapas de desarrollo y la restauración de la actividad telomerasa rescata la longitud de los telómeros y la supervivencia, indicando que la dosis de telomerasa es crucial. La inhibición genética de p53 rescata los efectos adversos de la perdidad de telómeros, indicando que los mecanismos moleculares están conservados desde peces hasta mamíferos. El estudio de regeneración demuestra que los genotipos tert+/+ y tert-/- regeneran después de una o varias escisiones, aunque la eficiencia de regeneración disminuye con el envejecimiento en ambos. La línea tert-/- regenera con un ratio de regeneración menor que tert+/+, y mantiene la longitud telomérica. ALT está implicado en el mantenimiento de los telómeros durante la regeneración de la aleta caudal en el pez mutante, y se necesitan más estudios para determinar el papel de la telomerasa y posiblemente de ALT en el pez cebra salvaje durante este proceso. - Objectives. Study of telomere length, telomerase expression and activity in zebrafish (Danio rerio) during the caudal fin regeneration process in wild-type and tert mutant - Methodology. For approaching the first goal, wild-type zebrafish from different strains and different ages have been used. The study of gene expression was carried out by RT-PCR and real time PCR. The telomerase activity was determined by using a specific and quantitative assay based in the amplification of the telomere sequence called Q-TRAP, and also analyzed quantitatively by TRAP. The telomere length was quantified using three different techniques: by a whole-mount in situ hybridization with a fluorescent telomeric probe (Q-FISH and Flow-FISH) or by hybridization using a radioactive telomeric probe (TRF assay). To address the second goal, three zebrafish genotypes: tert+/+, tert+/- y tert-/- have been used. Besides the above techniques, survival curves to estimate the mean lifespan of the first generation (G1) of tert mutant line, and a histological study of different tissues (Hematoxylin / Eosin, PAS and TUNEL staining) have been performed. To study the tert mutant line G2: survival curves, apoptosis by TUNEL assay, and Q-FISH on metaphasic nucleus. To achieve the third objective, we have used all three genotypes at different ages. Telomere length was analyzed by Flow-FISH, and the detection of telomeric DNA circles by CCassay. A caudal fin regeneration assay on adult with gene inhibition using morpholino following by electroporation was performed and finally a caudal fin regeneration assay on larvae with gene inhibition by immersion. Zebrafish were maintained as described in the zebrafish handbook. The experiments performed comply with the Guidelines of the European Union Council (86/609/EU). Experiments and procedures were performed as approved by the Bioethical Committee of the University Hospital "Virgen de la Arrixaca" (Spain) and by the Children's Hospital Boston institutional Animal Care and Use Committee (USA). - Results and conclusions. The results of this tesis have shown that the expression of telomerase and telomere length are closely related during the entire life cycle of the fish and that these two parameters can be used as biomarkers of aging in zebrafish. Telomerase-deficient zebrafish characterization have shown premature aging and reduced lifespan in the first generation, as occurs in humans and may be considered as a promising model to study telomere-driven aging. Interestingly, this mutant line reproduced the genetic anticipation phenomenon, related to telomere shortening, observed in humans with DC. The second-generation embryos died in early developmental stages, and restoration of telomerase activity rescued telomere length and survival, indicating that telomerase dosage is crucial. Genetic inhibition of p53 rescued the adverse effects of telomere loss, indicating that the molecular mechanisms induced by telomere shortening are conserved from fish to mammals. The regeneration study showed that all genotypes regenerated after single or successive amputations and that regeneration efficiency decreased with aging. Tert mutant line showed a lower regeneration rate than its wild-type counterpart, and maintained telomere length in the regenerative tissue. ALT mechanism is implicated in telomere maintenance during caudal fin regeneration in telomerase-deficient fish. Further studies are necessary to definitively elucidate the role of telomerase and the possible ALT implication in telomere maintenance during caudal fin regeneration in wild-type animals.

The importance of prudent and rational use of antimicrobial is important, not only to safeguard the efficacy of these drugs in humans and veterinary medicine but, even more so, to prevent the emergence and spread of undesirable resistance phenotypes in zoonotic pathogens as well as in commensal bacteria that can be transmitted between animals and humans. Even more importance and attention is now given to the prudent use of medically important antimicrobial drugs, referring to those drugs for human therapeutic use. The fluoroquinolones belong to this category. These are very potent antimicrobials and active against a wide range of pathogenic organisms and are well distributed in the body after administration. This class of antimicrobials has a therapeutic effect on most infections in different organs or tissues. Although it is rare that fluoroquinolones are the only available agent for treatment of a specific infectious disease, fluoroquinolones are important alternative medicinal products for a veterinarian to have as option for treatment. Fluoroquinolones have a unique mechanism of action not related to conventional antimicrobials, and therefore their efficacy should be retained as long as possible. The avian production increased enormously in the last 50 years and the European Union (EU) is one of the world's top producers in poultry meat and a net exporter of poultry products. In this production, turkeys that is considered a "minor species", but it is important in the livestock production of Italy. Scarce data exist about the usage of antimicrobial drugs in turkey and even less is known about their efficacy. As the limited number of medicinal products authorized in this species, antimicrobial therapy is frequently carried out with the few products authorized or with drugs "extra-label" used with the consequence of increases of selective pressure and also with the possibility of cross-resistance within the same pharmacological group of compounds. The studies reported in this thesis aimed to revise the use of fluoroquinolones in turkey to maintain the efficacy and reduce the spread of resistance against E.Coli, the most common zoonotic avian pathogen. Pharmacokinetic(PK)/pharmacodynamics (PD) models are the best tool in order to select optimal dosage regimen. To confirm dosages used at farms and allow the integration of PK/PD data, the plasma concentrations in blood from healthy animals collected during treatment with flumequine and enrofloxacin, were determined. The first step was to optimize and validate a fast, simple, sensitive, and specific liquid chromatography-mass spectrometry (LC-MS)/MS/MS method suitable for the detection of a wide range of concentrations of fluoroquinolones as those occurring in pharmacokinetic and residue depletion studies from several matrices. The first trial presents a sensitive and reliable confirmatory method for the extraction, identification, quantification of five fluoroquinolones . For the extraction and matrix clean-up of fluoroquinolones residues from all biological matrices, the Quick Easy Cheap Effective Rugged Safe (QuEChERS) methodology was adopted; only for plasma samples acetonitrile was used. The analyses were performed by (LC-MS. LC separation was performed on a C18 Kinetex column (100x2.1 mm, 2.6 µm, Phenomenex, CA, USA) with gradient elution using ammonium acetate solution (10 mM, pH 2.5) and methanol containing 0.1% formic acid. Mass spectrometric identification was done using an LTQ XL ion trap (Thermo Fisher Scientific, CA, USA), with a heated electrospray ionization probe, in positive ion mode. The method was validated according to the European Legislation (decision 2002/657/EC) and EMA guideline (EMA/CVMP/VICH/463202/2009); selectivity, linearity response, trueness (in terms of recovery), precision (within-day repeatability and within-laboratory reproducibility), limit of detection, limit of quantification, decision limits, detection capability, absolute recovery and robustness were evaluated using turkey blank matrices. All data were within the required limits established for confirmatory methods except for flumequine which presented a recovery value slightly higher than 110% in muscle and intestinal content. For all fluoroquinolones, all the extraction rates were greater than 70% and limits of quantification ranged from 1.2 µg/kg to 118.8 µg/kg. This method was suitable for the identification and quantification of fluoroquinolones in plasma samples of turkeys treated for the purpose of second and third trials. In the second trial, the PK behavior of flumequine administered to 32 healthy turkeys as an oral bolus via gavage or as 5 days of 10-hours pulsed administration in drinking water were compared, using the EU authorized dose of 15 mg/kg and the double dose of 30 mg/kg. The MIC of 235 Escherichia coli field strains isolated from poultry were determined for PD to develop a PK/PD model. Blood samples were collected at established times over 24 h, and the obtained plasma was analyzed using the LC-MS/MS/MS method previously described. A monocompartmental model and a noncompartmental model were applied to the data to obtain the PK results. The maximum concentration (Cmax)/MIC50 and the plasma concentration-time curve from 0 to 24 hours (AUC0–24)/MIC50 ratios were, respectively, 0.67 ± 0.09 and 4.76 ± 0.48 and 1.18 ± 0.35 and 7.05 ± 2.40 for the 15 and 30 mg/kg bolus doses, respectively. After 10-hours pulsed administration of 15 mg/kg, values of Cmax/MIC50, 0.19 ± 0.02 on day 1 and 0.30 ± 0.08 on day 5 of therapy were obtained, the AUC/MIC50 ratios were 2.09 ± 0.29 and 3.22 ± 0.93 on d 1 and 5, respectively. Higher values were obtained with the doubled dose of 30 mg/kg: the Cmax/MIC50 ratios were 0.49 ± 0.11 on day 1 and 0.69 ± 0.18 on day 5; the AUC/MIC50 ratios were 5.15 ± 1.15 and 6.57 ± 1.92 on d 1 and 5, respectively. For both types of administration and both dosages, the Cmax/MIC50 and the AUC/MIC50 ratios achieved were significantly lower than the fluoroquinolones breakpoints usually considered for efficacy. The last trial involving 50 healthy turkeys, was conducted to evaluate the efficacy of enrofloxacin. As in the previous study, the effectiveness of different treatment schemes against E. coli was evaluated by a PK/PD approach, correlating the PK results with the MIC determined for 235 E. coli strains. In this study, 3 different oral treatments (a single oral gavage, 5 days of 10-hours pulsed water medication, and 5 days of 24-hours continuous water medication) and single parenteral (subcutaneous; SC) treatment using 2 different doses of enrofloxacin (i.e., the EU authorized dose, 10 mg/kg, and double the EU recommended dose, 20 mg/kg) were evaluated. Blood samples were collected at established times over 24 h. Plasma was analyzed using a LC-MS/MS/MS that was validated in house. A monocompartmental and a noncompartmental model were applied to the data to obtain the PK results. After gavage administration, the mean maximum concentration Cmax/MIC50 and area under the curve AUC0–24/MIC50 ratios were, respectively, 3.07 ± 0.62 and 7.01 ± 1.03 and 25.48± 3.04 and 57.2 ± 3.73 for the 10 and 20 mg/kg doses, respectively. After SC administration of 10 mg/kg, Cmax/MIC50 and AUC0–24/MIC50 ratios were 3.45 ± 0.75 and 33.96 ± 7.46, respectively. After the administration of 10-h pulsed or 24-h continuous medicated water at 20 mg/kg, lower values of Cmax/MIC50 (10-h pulsed: 3.45 ± 0.7; 24-h continuous: 3.05 ±0.48) and AUC0–24/MIC50 (10-h pulsed: 42.42 ± 6.17; 24-h continuous: 53.32 ± 5.55) were obtained. Based on these results, the European Union-recommended dosage of 10 mg/kg seems ineffective to achieve adequate drug plasma concentrations and even the 20 mg/kg by 10 h pulsed or continuous medicated water administration did not reach completely efficacious concentrations in plasma against colibacillosis. Although the results obtained were not completely encouraging, the medicated water should preferably be provided continuously. To conclude about the efficacy of enrofloxacin treatment against colibacillosis, target tissue concentration should be extensively considered.
