Incorporating antioxidants in functional foods represents an excellent dietary approach to prevent oxidative stress. The present work aimed to create a novel formulation of biscuits using a combination of Zingiber cassumunar (ZC) and purple sweet potato (PSP) as the main ingredients, in producing a functional food product that possesses antioxidant properties. The present work involved conducting trials and implementing optimisation techniques in order to obtain three distinct biscuit formulations, namely F1 (ZC 0.75 g: PSP 5.25 g), F2 (ZC 0.45 g: PSP 5.55 g), and F3 (ZC 0.28 g: PSP 5.72 g). The proximate analysis of the three formulations met the Standard Nasional Indonesia (SNI) criteria for biscuits, which encompassed parameters such as moisture content, ash, fat, protein, crude fibre, and metal residue. The F3 biscuits were chosen as the test biscuits in subsequent experiments due to their superior quality. The administration of F3 biscuits at a dosage of 1.94 g per 200 g of body weight to rats fed with high-fat diet for 28 d resulted in a substantial reduction (p < 0.05) in malondialdehyde levels, with a mean value of 2.17 ± 0.22 nmol/mL. Additionally, this intervention improved superoxide dismutase activity, with a mean value of 72.95 ± 3.06%. The F3 biscuits demonstrated promising antioxidant potential. This could serve as a basis for further clinical trials in humans before its potential commercialisation. Further investigation into the formulation of the biscuit is necessary, especially concerning the moisture, ash, and crude fibre levels present.
18 Páginas.-- 5 Figuras.-- 3 Tablas ; The effect of nitrate/nitrite (0, 37.5, 75, and 150 mg/kg) in the dry-cured loin formulation on the formation of lipid and protein oxidation products during in vitro digestion was evaluated. Dry-cured loins formulated with nitrate/nitrite resulted in significantly less lipid and protein oxidation than uncured loins before and after simulated digestion. Compared to loins added with 0 mg/kg nitrate/nitrite, dry-cured loins with 37.5, 75, and 150 mg/kg contained a significantly lower content of conjugated dienes, malondialdehyde, carbonyls, and non-heme iron, and higher amounts of nitrosylmioglobin and thiols. During in vitro digestion, the content of conjugated dienes, malondialdehyde, and carbonyls increased, while thiol content decreased, indicating the development of lipid and protein oxidative processes. At the end of the intestinal phase, the 75 mg/kg digests had a significantly higher content of conjugated dienes, while no differences were found among the other digests. During the in vitro intestinal phase (180 and 240 min), nitrate/nitrite curing resulted in significantly lower malondialdehyde concentrations in the 37.5, 75, and 150 mg/kg loin digests than in the uncured loin digests. No significant differences were observed at the end of the intestinal digestion phase between the cured loin digests. Digests of dried loins without nitrate/nitrite addition showed higher carbonyl contents than the nitrate/nitrite cured counterparts. The loss of thiols was significantly higher in loin digests without added nitrate/nitrite than in loin digests with different amounts of curing salts. It is not known whether exposure to air pollutants causes systemic oxidative stress in children. We investigated the association between exposure to air pollution and biomarkers of oxidative stress in relation to a governmental air quality intervention implemented during the 2008 Beijing Olympic Games. We studied 36 schoolchildren during 5 time periods before and during the Olympic Games in Beijing (June 2007-September 2008). The oxidative stress biomarkers 8-oxo-7,8-dihydro-2'-deoxyguanosine (8-oxodG) and malondialdehyde were measured in urine samples collected daily during each period. Generalized estimating equations were used to examine the relationship between repeated biomarker measurements and ambient air pollutant levels. During the Olympic intervention period, substantial reductions in air pollution (-19% to -72%), urinary 8-oxodG concentrations (-37.4%; 95% confidence interval: -53.5, -15.7), and urinary malondialdehyde concentrations (-25.3%; 95% confidence interval: -34.3, -15.1) were found. Malondialdehyde and 8-oxodG were significantly associated with concentrations of black carbon, fine particulate matter with an aerodynamic with diameter less than 2.5 μm, sulfur dioxide, nitrogen dioxide, and carbon monoxide. Biomarker changes per each interquartile-range increase in pollutants were largest at lag 0 or lag 1. In a 2-pollutant model, the most robust associations were for black carbon. These findings suggest that exposure to black carbon leads to systemic oxidative stress in children.
[EN] Little information is available on the physiological and biochemical responses to water stress in eggplant (Solanum melongena). We valuated four genetically diverse eggplant varieties (MEL3-MEL6) under control and water stress conditions. Measurements were taken for plant growth, tissue water content, levels of chlorophylls a and b, carotenoids, roline, malondialdehyde, total phenolics, total flavonoids, superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase APX), and glutathione reductase (GR) activities. For most traits, the water stress treatment had a greater contribution than the variety effect to the total sums of squares in an ANOVA analysis, except for total flavonoids, SOD, APX, and GR. The water stress treatment had a strong effect on plant growth and tissue water content. In general, water tress reduced the three photosynthetic pigments, increased proline, malondialdehyde, total phenolics, and total flavonoids, although some varietal differences were ob- served. Different patterns were also detected in the activities of the four enzymes evaluated, but few differences were observed for individual varieties between the control and water stress treatments. Many significant phe- notypic orrelations were observed among the traits studied, but only eight environmental correlations were detected. A PCA analysis distinctly separated individuals according to the treatment, and revealed a clearer separation of varieties under water stress than under control conditions, pointing to varietal differences in the responses to stress. Our results suggest that proline could be used as a marker for drought stress tolerance in this species. Perilla frutescens L. gehört in die Familie der Lippenblütler, stammt ursprünglich aus Asien, wo sie seit Jahrhunderten traditionell als Heil- und Gewürzpflanze genutzt wird. Inzwischen wird sie auch in der Europäischen Union kommerziell genutzt und hier u.a. als Nahrungsergänzungsmittel, als Zusatz in Kosmetika und als homöopathisches Arzneimittel eingesetzt. Der Rohstoff wird aus Asien importiert. Die Gattung Perilla umfasst zahlreiche Arten, die sich in Gehalt, Zusammensetzung und antioxidativen Eigenschaften von Carotinoiden und Polyphenolen signifikant unterscheiden. Einige Arten enthalten das toxisch wirkende Perillaketon als Bestandteil des etherischen Öls. In den ABTS- und LPO Testsystemen zeigten Extrakte der untersuchten Genotypen insgesamt sehr gute antioxidative Eigenschaften und signifikante Unterschiede zwischen den Genotypen. Im Blutplasma von Diabetikern und gesunden Probanden wurde nach einer zwei-wöchigen Supplementierung mit einem Perilla-Präparat eine signifikant höhere Luteinkonzentration gemessen. Die Konzentration von Malondialdehyd verringerte sich in Gesunden tendenziell, in Diabetikern signifikant. Generell zeigten alle untersuchten Genotypen sehr gute Anbau- und Ertragseigenschaften und eignen sich für den kommerziellen Anbau im Freiland. ; Perilla frutescens belongs to the Lamiaceae and is a medical and aromatic plant originating from Asia, where it is used traditionally since millennia. Meanwhile it is also commercially used in the European Union as a dietary supplement, additive in cosmetics and as a homeopathic remedy. The raw material is imported from Asia. The genus Perilla encompasses a variety of species that significantly differ in content, composition and antioxidative capacity of carotenoids and polyphenols. Some of the species contain the toxic perillaketone as a component of the essential oil. Generally the extracts of the analysed genotypes showed very high antioxidative capacities in the ABTS- and the LPO-test systems with significant differences between the tested genotypes. After a two-week supplementation with a Perilla preparation, the concentration of lutein was significantly higher in blood-plasma of either diabetic and healthy volunteers. The concentration of malondialdehyde decreased in healthy volunteers by trend and significantly in diabetic test persons. Generally all of the analysed genotypes indicated very good cultivation characteristics and are suitable for a commercial cultivation in the open field.
This study was designed to investigate the protective effects of red cabbage and garlic extracts on the oxidative stress induced through treatment with fumonisin B1 in male mice. The study was conducted on sixty male mice that were divided randomly into six groups. Group 1included control mice, mice in group 2 received a daily oral dose (200 µg/kg.b.w) of fumonisin B1 for one month to induced mycotoxicosis, mice in group 3 received 500 mg/kg.b.w red cabbage extract plus fumonisin B1, mice in group 4 received only red cabbage extract, mice in group 5 received 500 mg/kg.b.w garlic extract plus fumonisin B1, and group 6 received garlic extract alone. After finishing the experiments, samples of blood were used for biochemical examination. The results indicated that group 2 mice had significantly increased (p < 0.05) serum levels of malondialdehyde, which is a biomarker of lipid peroxidation, in addition to significantly decreased levels of catalase and the antioxidant enzymes glutathione peroxidase , glutathione , and superoxide dismutase , as compared to the normal control group. Oral administration of garlic and red cabbage extracts produced significantly decreased levels of serum malondialdehyde and significantly increased levels of superoxide dismutase, catalase, glutathione peroxidase, and glutathione, as compared to group 2. These results indicated that the experimental treatment with fumonisin B-1 caused an elevated oxidative stress in addition to the consumption of the antioxidant enzymes in response to the induced oxidative aggression.
